First off, I'd recommend some books about road trips through Tuscany. They'll give you some great ideas. My tips will probably be more about the places I've seen myself, but it's a good way to start. Most of the authors aren't from Tuscany, but the region's attracted a lot of talented people from everywhere, so it's all good.
